揭開頭孢吡普穩(wěn)定性的秘密—雙技術(shù)聯(lián)手解碼降解機(jī)制

頭孢吡普作為一種新型的β-內(nèi)酰胺類抗生素，對(duì)耐甲氧西林金黃色葡萄球菌、耐萬古霉素金黃色葡萄球菌和耐青霉素肺炎鏈球菌均具有活性。為了產(chǎn)生可能在相關(guān)儲(chǔ)存條件下形成的頭孢吡普潛在降解產(chǎn)物（DPs），在溶液和固態(tài)下分別進(jìn)行了酸性、堿性、氧化、光解和熱解強(qiáng)制降解試驗(yàn)。

采用Kinetex Biphenyl色譜柱，在pH 5.8的醋酸銨緩沖液和乙腈體系下，開發(fā)了一種高選擇性的HPLC分析方法，用于分離頭孢吡普及其DPs和合成副產(chǎn)物（SBPs）。并基于質(zhì)譜分析，提出了13種新的SBPs和DPs的結(jié)構(gòu)式，以及在溶液中發(fā)現(xiàn)的另外三種DPs的分子式和四種固態(tài)降解下的氧化DPs的分子式。
